Neurodegenerative disorders are a highly prevalent class of diseases, whose pathological mechanisms start before the appearance of any clear symptoms. This fact has prompted scientists to search for biomarkers that could aid early treatment. These currently incurable pathologies share the presence of aberrant aggregates called amyloids in the nervous system, which are composed of specific proteins. In this review, we discuss how these proteins, their conformations and modifications could be exploited as biomarkers for diagnostic purposes. We focus on proteins that are associated with the most prevalent neurodegenerative disorders, including Alzheimer's and Parkinson's diseases, amyotrophic lateral sclerosis, and frontotemporal dementia. We also describe current challenges in detection, the most recent techniques with diagnostic potentials and possible future developments in diagnosis.A comparative study of the possibilities of using ribokinase → phosphopentomutase → nucleoside phosphorylase cascades in the synthesis of modified nucleosides was carried out. Recombinant phosphopentomutase from Thermus thermophilus HB27 was obtained for the first time a strain producing a soluble form of the enzyme was created, and a method for its isolation and chromatographic purification was developed. It was shown that cascade syntheses of modified nucleosides can be carried out both by the mesophilic and thermophilic routes from D-pentoses ribose, 2-deoxyribose, arabinose, xylose, and 2-deoxy-2-fluoroarabinose. The efficiency of 2-chloradenine nucleoside synthesis decreases in the following order Rib (92), dRib (74), Ara (66), F-Ara (8), and Xyl (2%) in 30 min for mesophilic enzymes. For thermophilic enzymes Rib (76), dRib (62), Ara (32), F-Ara ( less then 1), and Xyl (2%) in 30 min. Upon incubation of the reaction mixtures for a day, the amounts of 2-chloroadenine riboside (thermophilic cascade), 2-deoxyribosides (both cascades), and arabinoside (mesophilic cascade) decreased roughly by half. The conversion of the base to 2-fluoroarabinosides and xylosides continued to increase in both cases and reached 20-40%. Four nucleosides were quantitatively produced by a cascade of enzymes from D-ribose and D-arabinose. The ribosides of 8-azaguanine (thermophilic cascade) and allopurinol (mesophilic cascade) were synthesized. For the first time, D-arabinosides of 2-chloro-6-methoxypurine and 2-fluoro-6-methoxypurine were synthesized using the mesophilic cascade. Despite the relatively small difference in temperatures when performing the cascade reactions (50 and 80 °C), the rate of product formation in the reactions with Escherichia coli enzymes was significantly higher. E. coli enzymes also provided a higher content of the target products in the reaction mixture. Therefore, they are more appropriate for use in the polyenzymatic synthesis of modified nucleosides.Infrared thermography has been widely adopted in many applications for material structure inspection, where data analysis methods are often implemented to elaborate raw thermal data and to characterize material structural properties. Herein, a multiscale thermographic data analysis framework is proposed and applied to building structure inspection. In detail, thermograms are first collected by conducting solar loading thermography, which are then decomposed into several intrinsic mode functions under different spatial scales by multidimensional ensemble empirical mode decomposition. At each scale, principal component analysis (PCA) is implemented for feature extraction. Compared with principal component thermography that applies PCA directly to raw thermal data, the proposed multiscale analysis method is able to zoom in on different types of structural features.In order to cope with recent climate change, Korea is reducing the use of heavy oil in petroleum-fired power plants and mixing bio-oils. Accordingly, this must be taken into account when calculating the emissions of air pollutants. However, in the case of Korea, when calculating NH3 emissions, the United States Environmental Protection Agency (EPA) emission factor is applied as it is to calculate emissions, and for petroleum power plants, the heavy oil emission factor proposed by EPA is used as it is to calculate emissions. In petroleum power plants, bio-oil is not mixed in a certain amount and used at a different ratio depending on the situation of the power plant. Therefore, in this study, the NH3 emission factor according to the mixing ratio of bio-heavy oil is calculated and the mixing ratio is calculated. As a result of the analysis, the emission factor according to bio-oil and the mixed ratio was found to be in the range of 0.010~0.033 kg NH3/kL, and it was lower than the heavy oil emission factor 0.096 kg NH3/kL of EPA currently used in Korea. This is because the amount of NH3 through the slip is also small since the use of NH3 for reduction is also low because the NOx emission from the use of bio-oil is low. Considering all of these points, we have statistically analyzed whether emission factors should be developed and applied. As a result of the confirmation, the difference according to the mixed consumption rate was not large.
Medial open wedge high tibial osteotomy (MOWHTO) is a well-established treatment for osteoarthritis of the medial tibiofemoral compartment. Surgical site infection (SSI) after MOWHTO is a devastating complication that may require further surgery. In this study, we aimed to identify the risk factors for infection after MOWHTO over 1 to 4 years of follow-up.
Fifty-nine patients who underwent MOWHTO combined with knee arthroscopic surgery were included in this prospective study. Artificial bone grafts were used in all cases. Possible risk factors, including sex, age, body mass index (BMI), underlying disease, hospitalization length, correction angle, and surgery time, were recorded. Both univariate and multivariate analysis were used.
A total of 59 patients who underwent 61 operations were included. Eleven patients (18.0%) were reported to have SSI. Univariate analysis showed that smoking and diabetes mellitus were positively associated with SSI. Multivariate analysis showed that smoking and age were positively associated with SSI.
